Viral capsid of Anellovirus and ssDNA reporter constructs carrying an anellovirus intergenic region were both produced separately in E. coli systems, purified and then mixed together in an in vitro system. At particular molar ratios we noted that the ssDNA was spontaneously encapsulated into anellovirus capsids. Viral vector structures were visualized by TEM and quantified by qPCR suggesting reasonable and scalable production efficiency.en
